The Landscape of DNA Methylation Generates Insight Into Epigenetic Regulation of Differences Between Slow-Twitch and Fast-Twitch Muscles in Pseudocaranx dentex

Skeletal muscles of teleost are mainly composed slow-twitch (SM) and fast-twitch (FM) differed in contractile properties, metabolic capacities, regeneration rate. The transcriptional regulatory mechanisms that control different muscle types have been elucidated according to transcriptome between SM FM. However, the differences FM were affected not only by genotype but also complicated epigenetic effects, including DNA methylation, which usually regulates genes transcription level. To determine essential role methylation regulation types, we analyzed whole-genome profiles pelagic migratory fish Pseudocaranx dentex with abundant well-separated integrated previously obtained data. A total 4,217 differentially methylated (DMGs) identified, 3,582 located gene body 635 promoter. These DMGs participated metabolite cell junction. Enriched junction pathway reflected capillary distribution Through comprehensive analysis methylome transcriptome, 84 expressed (DEGs) showed significant variation promoters FM, indicating their expression was regulated methylation. Hypomethylated highly oxygen storage protein Myoglobin ( myg ) indicated demethylation promoter could upregulate its expression, thus increasing O 2 supplying meeting demands SM. Hypermethylated lowly tnn (Troponin) rlc (myosin light chain) may be associated low mobility myosin cross bridges, lead slower less frequent contraction than In addition, hypomethylated lbx1 (Ladybird homeobox homolog 1) epo (erythropoietin) related increased satellite numbers, Semaphorin/Plexin higher rate neuromuscular connection reconstruction, further promote high efficiency Our study potential regulate physiological characteristics facilitate our understanding skeletal adaptation fishes enrich theoretical basis for adaptive evolution fishes.
